Is the connection to Systems Manager via a VPC endpoint?
In that case, please make sure that the three VPC endpoints described in the document below have been created.

Yes, VPC endpoints are available and able to connect to them using traceroute and nc tool I ran these test. It's working with EC2 --> Connect --> Session manager -> Connect. But issue is not showing up in fleet manager. We have windows machine, so we need RDP connection to Ec2 using fleet manager.
