No. Documentation states that database with replication allows the proxy to connect to the writer instance only.
Also the proxy needs to be in the same VPC as the RDS instance.

This is possible with Aurora. Your questions is tagged with MySQL so if that's what you're interested in, it doesn't look like this functionality is available (yet).
